Interested in how many rounds of Carfilzomib (combo or single) it took before you started to have a response and what kind of response you ultimately achieved, as in VGPR, CR, etc..
Very interested to know if anyone had increased M Spike during early rounds of Kyprolis (carfilzomib) and had the M Spike decrease in later rounds.

When my husband was first diagnosed almost 3 years ago, he had numerous lesions, anemia, and his M spike was either 5.1 or 4.9 depending on the lab. He had one bone marrow biopsy (BMB) from the initial oncologist who diagnosed him, and a second about a month later when he first saw the myeloma specialists at the University of Michigan, but had had no treatment yet.
He began a clinical study taking carfilzomib [Kyprolis], Revlimid and dexamethasone and was on the combination for a total of 24 months. His M spike dropped dramatically the first few months and he was VGPR by the fourth month. Around the sixth month, his M spike continued to drop, but more slowly. From approximately 18 months to 24 months, it stayed at 0.1.
He was then put on just Revlimid and, finally about 3 months after that, he reached 0.0 and has remained there since then. He had another BMB and flow cytometry and neither one showed any monoclonal plasma cells. He also has had a normal FLC ratio, so I think he is considered to have a SCR.
He never had any increase in M spike, but the decrease just seemed to get smaller at a snail's pace for such a long time. As his treatment continued, we were told not to expect such dramatic decreases as he had at first, and that even in the 23 and 24th month it could still drop.

Yes, you are correct. I am not responding yet to Round 3 of CCd. Do you have any related data as to when Carfilzomib "kicks" in, if ever?

Is this your first treatment for myeloma? If so, the data discussed in the abstract should be particularly helpful to you. If you are being treated for a relapse, then the data from the presentation should be intrepreted more carefully.
