There is a lot of data online if you look up Dr. Jakubowiak and CRD, but here are the study results for those who asked:
• Carfilzomib (Kyprolis) is a next-generation proteasome inhibitor that selectively and irreversibly binds to its target, resulting in sustained inhibition in the absence of off-target effects.
• In relapsed and/or refractory multiple myeloma, the combination of carfilzomib (CFZ) with lenalidomide (Len) and low-dose dexamethasone (Dex) (CRd) has shown very promising efficacy: 78% ≥PR , 40% ≥VGPR, 24% CR/nCR (Wang et al, ASCO 2011).
• In a Phase I/II study of newly diagnosed multiple myeloma, the regimen was well tolerated and very active with 96% ≥PR, 70% ≥VGPR and 55% CR/nCR (Jakubowiak et al, ASH 2010).
• This study presents results after enrollment in the Phase II portion of the Phase I/II trial of CRd in multiple myeloma.
We are now thinking my numbers went up because I needed a higher dose of Carfilzomib than the study allowed. I am on the increased dose now and things have quickly turned around.

my creatinine has been normal before carfilzomid.it started to creep up after first cycle.the 2nd cycle with increasing the dose,it has doubled and in a zone where they couldn`t give my infusion today.has anyone had this happen

Hi Al, I did carfilzomib/rev/dex for 8 cycles and my creatinine stayed normal. Are you on Zometa or Aredia?.....sometimes they affect the kidneys. Good luck. I hope they figure it out as carfilzomib has really worked for me and others in my trial.

i have been getting aredia since 2002,never had a problem
they gave me aredia first then 500mg of hydration then 50 mg. of carfilzimid then 500 mg hydration after.
if it`s because of either drug is it permanent the damage

My daughter started on carfilzomib / dex 2 months ago. So far, no side effects, except the dex messes up her sleep pattern. The first night she can't sleep at all. By the fourth day she crashes.

Is your daughter having only Kyprolis plus dex? Many people here are having additional drug like Revlimid.
My mother is also having Kyprolis plus dex as she couldn't tolerate Revlimid and Velcade. She just finished her 1st cycle and so far no side effects.
I am trying to find out if this combination of Kyprolis plus dex is effective.

Today I start Kyprolis (carfilzomib), Revlimid and dex. I'll report what kind of side effects I have.
I completed 6 cycles of Velcade, Revlimid, and dex. At first, I had a great response - my tumors all melted away! But six weeks later, they were all back, plus some new ones. This was crushing news. I only got four months out of RVD.
So yesterday I had a power port placed, and today is my first dose of carfilzomib.
I'm hoping to get back to a point where I can have an allogeneic transplant, and I'm hoping to find a match. I was worried that this relapse would change the risk / benefit calculation enough to make allo an unreasonable option. However, Dr. Laubach at DFCI is still recommending an allo. And, honestly, for someone my age, even if the risk / benefit calculation regarding allo does change some, what's the alternative anyway?!?!

I will certainly follow your posts about this treatment regularly. Also hope you soon find the donor for your allo.
I sincerely hope you won't need any transplant at all, by achieving a complete remission with this new treatment.
