My FISH showed 10% of plasma cells with deletion 8. My question is did the cancer cause the deletion or did I have it before the cancer and it may be one of the reasons I have cancer?
I haven't heard of any one else mentioning they have this deletion, thanks.

Re: Deletion of chromosome 8
Hi Dee,
I'm going to guess that your FISH actually said an "8p" deletion, not just "8" (the 8 means chromosome 8 and the p means the short arm of that chromosome)
There are all kinds of mutations associated with myeloma...and then there are those mutations that have been identified as having an impact on prognosis. You obviously hear about the latter, because those are the ones that either scare us or makes us feel relieved if they are good indicators. Then there are those that just don't matter.
8p deletions are pretty common in myeloma, but are not identified as having prognostic significance (that is, they don't matter).

BTW, would you mind sharing your most recent numbers with us in comparison to your time of starting your unique treatment? I am amazingly curious as to how your treatment is impacting your numbers.

Re: Deletion of chromosome 8
In this study,
http://bloodjournal.hematologylibrary.org/content/116/15/e56.full
chromosome 8 deletions were detected in 25% of the patient samples evaluated by FISH, and a subset of those also was associated with increased expression of c-myc, an oncogene.
In this Myeloma Beacon report,
https://myelomabeacon.org/news/2013/03/06/beacon-newsflashes-march-6-2013/
there was discussion of an abstract by a French group with similar percentages of patients with deletions in chromosome 8. Here, it was postulated that the deletion might result in lack or lowered expression of the TRAIL family receptors, which are involved in cell death. Disruption of this pathway may lend a hand to oncogenesis.
I didn't see information anywhere discussing prognosis of myeloma patients with this deletion.

Re: Deletion of chromosome 8
Thank you, it does say loss of chromosome 8. I can't find much information on it either and was just curious. I will look up the links.
Multibilly, I am having a physical done with my primary Feb. 7th. He is running blood work that day so I will share when I get the results back, I am very eager myself as to how the protocol is working. I came down with a bad flu that caused inflammation of my lungs and bronchial but they checked my oxygen and it showed 99% which is good, so I must have a bit of red blood cells circulating. My primary walked in and couldn't get over how good I looked even with the flu. LOL! I do not have an oncologist as my last one said if I didn't do treatment he wouldn't see me, he called a Saturday morning even though I had already canceled my appointments knowing that. I don't blame him as he was sure I would be dead, I am sure he reads the obituary regularly, LOL! So I will update later. Thank you.

Re: Deletion of chromosome 8
Glad you are doing so much better Dee! Things sounded dire for you a year or so ago.

Re: Deletion of chromosome 8
Yes! My kidneys were failing. The creat, is now .08 with GFRs in the 90 according my last lab in October. I am even back to playing my flute in front of a congregation! I couldn't do that before because of the anemia. We'll see what the difference is since October 11th at the beginning of my protocol on February 7th.
Re: Deletion of chromosome 8
Looking forward to the results. Take care.

Re: Deletion of chromosome 8
Hi,
I was just recently found to hit the smoldering myeloma mark of 10% plasma cell population in bone marrow and i found this thread searching for chromosome 8 deletion as on my recent bone marrow biopsy report it was noted that :
"cytogenic on bone marrow revealed deletion of chromosome 8q"
Searching around I've found that it seems it is common for 8p (short arm) to be found deleted in myeloma patients but I'm not finding anything related to the deletion of 8q (long arm).
So i'm not sure if the deletion of 8q ties into my smoldering myeloma at all or not.
Also the partial marrow results indicate aberrant plasma cells positive for CD56 and CD117 (not sure yet as to the meaning of this as well but searching around it seems hyperdiploidy comes up often as well as myeloma).
My full marrow results should be available in a few days and I'll have lots of questions for my treatment team at that time I'm sure.

Re: Deletion of chromosome 8
Well, 3 years later, and the 8q deletion hasn't changed or seemed to have mattered as of yet.
Since 2014 my plasma cell marrow count hasn't risen above 10%, but my M-spike did increase a little and my free light chain ratio is above normal as well as an overall high kappa level (matching IgG kappa clonal cells).
I've also developed a measurable amount of Bence-Jones proteinuria for the past year.
Oh, and immunoparesis set in a couple of years ago (low IgM).
So I'm overall healthy (sans for a five-month old staph aureus infection that is slowly finally clearing), but I do have a few risk factors for possible progression:
Three years later I haven't linked the del 8q details in my previous post with plasma cell dyscrasia. Hopefully in three more years I'll still be fine even if additional risk factors such as the items above continue to appear.

- 10% bone marrow plasma cell percentage (high-risk MGUS /smoldering myeloma range)
- Abnormal free-light chain ratio
