counting cell's in aspirate and biopsy

What is the standard practice for estimating plasma cell load in a biopsy or aspirate? I was given a range yesterday at my appointment and am a little confused. The range of my plasma cell load was 14-30%. I assume that is based on technique, no stain, visual count, with stain...
So which is usually the most accurate method? Do most institutions use the same method?
