Selinexor + dexamethasone - initial trial results

There was a press release put out earlier today by Karyopharm, the company that is developing the potential new myeloma therapy known as Selinexor. This drug also has been known in the past at KPT-330. The press release discusses updated results from a very small trial (8 patients thus far) testing Selinexor in combination with dexamethasone in relapsed myeloma patients.

Patients in the study had a median of 5.5 previous therapies. The overall response rate to the combination is 50 percent thus far. The clinical benefit rate, which is the overall response rate plus the percent of patients achieving a minor response or stable disease, is 75 percent.

One patient achieved a stringent complete response.

These are promising results. You don't often see an overall response rate of 50 percent in relapsed myeloma patients with so many previous lines of therapy.

That said, only eight patients have been tested thus far with the Selinexor-dex combination, so it's too early to draw any firm conclusions about how active this combination is.

There were two Selinexor abstracts related to myeloma presented at the ASH 2013 meeting in December. They were:

To give a sense of the impact this news had yesterday, the stock of Karyopharm, the company developing Selinexor, went up over 90 percent in price yesterday. The stock closed on Thursday afternoon at a price of about $25, and, by Friday afternoon at the closing bell, it was at $47 per share.

The market value of the company changed from about $750 million to $1.4 billion in one day.

My mother is going on a trial of selinexor-dex for quad-refractory myeloma. Hopefully her results will be just as good as this first cohort!

Thank you TerryH! We're about ready for some success, I can tell you that! :) Mom will have another drain of her myelomatous ascites on Monday and hopefully start the trial that day. Obviously our first hope is that it works at all, but also that it will work for a while and that the side effects are tolerable. I will keep the forum posted!
