I just started my 2nd cycle with carfilzomib (Krypolis), Revlimid, and dex. The first cycle I really didn't notice much in the way of side effects, but at the time I was feeling pretty poorly from the myeloma itself, so it's difficult to separate the two.

Now, after the first week of the 2nd cycle, I can say that I still think the side effects are overall less than with bortezomib (Velcade), but I have noticed some shortness of breath. It's pretty mild and really does not interfere with my function at all. I'm still exercising and walking up and down my stairs.

Looking back to the first cycle, I think I had the same mild shortness of breath then, too, but it was eclipsed by the myeloma side effects.

I mentioned to my nurse yesterday that I though the side effects were better with carfilzomib than with bortezomib, and she said, "You know, people tend to think bortezomib is benign because it's just a quick subcutaneous injections, but it's really not that benign. People do get serious side effects from it."

Hi Dano.

I have been on Kyprolis / dex for over a year now. I do experience fatigue for 2 or 3 days after infusion and, the night of my infusion, a headache. However, I believe I also have the side effects from the dex.

All in all, I have so far had a pretty good experience with Kyprolis. I have had no nausea or neurop­athy, which I had with Velcade, or a bad skin reaction, which I had with Revlimid.

Thanks to everyone for sharing their experience with Kyprolis.

I just completed my first three-week cycle with Kyprolis and dex, and my only side effect has been some minor fatigue. So far, my blood counts have remained normal or near normal, and I'm also tolerating the dex better than I have in the past. I get IV fluids (normal saline) before and after the Kyprolis infusion. I understand that to be the standard practice.

I also learned recently that getting one infusion at double the dose is just as effective as the two-day infusion. Has anyone had this regimen? It sure would make it more convenient.

I will have two more rounds of Kyprolis / dex before I have myeloma labs done in late April.
